Microscopy Giardia lamblia
this picture shows multiple views of single giardia lamblia (intestinalis) cyst imaged @ different instrument settings confocal microscopy. bar = 10 micrometers
(a) cyst imaged transmission (differential interference contrast), only.
(b) cyst wall selectively imaged through use of fluorescent-labelled (tritc) antibody cyst-wall specific.
(c) cyst imaged through use of carboxy fluorescein diacetate, viability stain.
(d) composite image of (b) , (c).
(e) composite image of (a), (b), , (c).
under normal compound light microscope, giardia looks clown face , 2 nuclei outlined adhesive discs above dark median bodies form mouth . cysts oval, have 4 nuclei, , have visible axostyles.
most eukaryotes have mitochondria, though giardia 1 of few lack these, contain mitochondrial relics, called mitosomes. such organism mixotricha paradoxa. however, similar other eukaryotes, giardia contains complex endomembrane system tubulin based cytoskeleton. immunoelectron microscopy antibody alpha-tubulin shows microtubules present in giardia in different structures such axonemes, basal bodies, adhesive discs, funis , median bodies.
